<p>Phycotoxins produced by toxinogenic dinoflagellates pose serious risks to bivalve aquaculture and seafood safety. This study assessed the occurrence of Diarrheic Shellfish Poisoning (DSP) toxins in <i>Perna perna</i> mussels cultivated in Arraial do Cabo, Rio de Janeiro, from January 2013 to May 2014 through both mouse bioassays and chromatographic analyses. Dinophysistoxins (DTX-1 and DTX-3), associated with okadaic acid (OA) group toxins, were confirmed by HPLC/MS in mussel hepatopancreas. DSP-positive events occurred in January, March, and May 2014, coinciding with the detection of <i>Dinophysis</i> spp. cells in water samples. Phycotoxins dinophysistoxin 1 and 3 were confirmed through a chromatographic analysis (HPLC/MS). This is the first historical record of DSP toxins in <i>P. perna</i> from Arraial do Cabo, an area of high ecological value and intense tourism. The findings highlight the vulnerability of local mariculture to HABs and reinforce the need for continuous phycotoxin monitoring to safeguard public health, sustain fisheries, and support the socioeconomic relevance of mussel farming in southeastern Brazil.</p>
