Therapeutic Effects of 6-Gingerol in Nerve Regeneration Following Sciatic Nerve Crush Injury: A Rat Model Study

Purpose

This study assesses the effects of 6-gingerol treatment on nerve regeneration in male Wistar rats following sciatic nerve crushing.

Method

To induce injury, rats were subjected to a crush injury. Intraperitoneal administration of either 6-Gingerol or a vehicle was carried out daily. Treatment groups received 6-Gingerol at doses of 10 or 20 mg/kg. The control group received the vehicle. The behavioral, electrophysiological, morphological, and ELISA parameters were evaluated.

Results

After surgery, rats displayed significant motor and sensory deficits. However, the rats that received 6-gingerol treatment (20 mg/kg) showed remarkable improvements in their sciatic functional index (SFI) and withdrawal reflex latency (WRL) values compared to the crush group. This suggests an improvement in both sensory and motor functions. Additionally, the group that received a combination of crush injury and 6-gingerol (20 mg/kg) showed a significant increase in nerve conduction velocity (NCV) and compound muscle action potential (CMAP) amplitude compared to the crush group. This indicates that the 6-gingerol treatment enhanced the axonal regeneration. Furthermore, histomorphometric analysis revealed a significant increase in myelinated fiber number in the crush/6-gingerol (20 mg/kg) group. The ELISA results also showed a reduction in the pro-inflammatory cytokines (IL-1β and TNF-α) and an increase in anti-inflammatory cytokines (IL-4 and IL-10) in the group that received a combination of crush injury and 6-gingerol (20 mg/kg) compared to the crush and crush/6-gingerol (10 mg/kg) groups.

Conclusion

These findings suggest that 6-gingerol treatment, particularly at the 20 mg/kg dosage, holds promise for promoting nerve regeneration and modulating inflammatory responses.
