<p>This study explored the diversity and pathogenicity of <i>Macrophomina</i> spp. in watermelon-producing regions of Ceará and Rio Grande do Norte States, Brazil. From 16 commercial fields across both states, we collected 30 watermelon plants exhibiting "root rot and vine decline" (RRVD) symptoms. Fungal growth was identified from root isolates based on morphological characteristics. The species of each isolate were further determined using specific primers: MpTefF and MpTefR for <i>M. phaseolina</i>, MsTefF and MsTefR for <i>M. pseudophaseolina</i>, and MeTefF and MeTefR for <i>M. euphorbiicola</i>. Out of 156 confirmed <i>Macrophomina</i> isolates, 75 were identified as <i>M. phaseolina</i> and 81 as <i>M. pseudophaseolina</i>. We assessed the disease incidence and severity of 50 isolates from each species on the watermelon variety 'Crimson Sweet' using infested husk rice grain inoculation. The parameters of shoot and root length, and dry weight of all plants were also evaluated. All isolates proved pathogenic to watermelon. Notably, plants inoculated with <i>M. phaseolina</i> showed greater disease incidence and severity than those colonized by <i>M. pseudophaseolina</i>. Moreover, <i>M. phaseolina</i>-inoculated plants had longer average shoot and root lengths but lower dry weight. In conclusion, our findings present the first report on the pathogenicity of <i>M. pseudophaseolina</i> to watermelon plants and highlight its predominant presence in the surveyed regions. Still, <i>M. phaseolina</i> exhibited greater severity toward watermelon plants.</p>
