Analysis of Yeast Extract Components Important for the Activity of Alicyclobacillus sp. SO9

While the presence of chloride ions strongly inhibits most bioleaching bacteria, the newly isolated acidophilic, iron-oxidizing bacterium Alicyclobacillus sp. SO9 even requires chloride. The strain has so far been maintained with different electron donors, but always in the presence of 0.02% yeast extract. To obtain a better understanding of the unusual traits of the strain we were interested in the precise role that yeast extract components play for Alicyclobacillus sp. SO9. The present study demonstrates the application of a novel strategy, based on oxygen consumption, for bacterial activity assessment using PyroScience® Oxygen Sensor Spots. The investigation revealed that thiamine and niacin are the most important vitamins for respiration of Alicyclobacillus sp. SO9 with glucose. In addition, some amino acids from the aspartate family (Asp, Met, Thr, Lys) may be required. Potassium had a positive effect on respiratory activity, but its addition to the two vitamins and casamino acids was not sufficient to replace yeast extract. Ultra- and nano-filtration experiments revealed that the missing compounds should have a mass smaller than 1 kDa and some also smaller than 0.5 kDa. ESI-FT-ICR-MS analyses of yeast extract components bound to an ENV + column revealed numerous signals, probably due to an enrichment with a bias for hydrophobic compounds, that are unlikely to play a role for SO9. However, some more polar compounds seem to be taken up and further analyses may identify some of the missing compounds.
