Insomnia and the Orexinergic Pathway in the Link with Psychopathology: Effects of DORAs in Insomnia Comorbid with Mental Disturbances

Insomnia can act as a precipitating factor, a comorbid disorder, or a transdiagnostic element for many mental disorders. The stress system’s hyperactivation has been identified as the key factor in insomnia that contributes to mental disorders. It is possible that insomnia may utilize the orexinergic pathway as a link with psychopathology. Thus, this review is aimed at discussing how the orexinergic pathway may play a role in insomnia and psychopathology. We reviewed all available data regarding the effect of DORAs on insomnia and comorbid mental disturbances in a systematic manner.
Recent FindingsPRISMA guidelines have led to the selection of 19 studies on the use of DORAs in preclinical, experimental, and clinical studies including, sleep, insomnia, and mental conditions.
SummaryAlthough further investigation is needed, targeting insomnia by using DORAs can be considered a promising therapeutic approach for psychiatric disorders.
