<p>The present study assesses variation in pollen viability and germinability among ten cultivars of Indian mango (<i>Mangifera indica</i> L.) in support of traditional breeding, germplasm conservation, and biotechnology applications. Pollen viability and germinability varied significantly depending on cultivar, flower type, storage conditions, and test method. Pollen from hermaphrodite flowers had higher viabilities than that of male flowers when assessed by either fluorescein diacetate (FDA) staining or in vitro germination, with t-tests revealing significant differences between hermaphrodites and males in nine of ten and seven of ten cultivars, respectively. Overall, FDA staining gave higher pollen viabilities than in vitro germination, with in vitro rates being the highest at 35&#xa0;℃ in culture media containing 40% sucrose. Relative differences in viability among cultivars were greatest under standard culture conditions (35&#xa0;℃, 40% sucrose), with ‘Beneshan’ and ‘Neelum’ having comparatively high viabilities, ‘Dasheri’, ‘Proddaturavakaya’, and ‘Rumani’ having intermediate viabilities, and Khadher, Kesar, KMH1, ‘Prabhasankar’, and Pulihora having low viabilities. Storage of FDA-tested pollen for 48&#xa0;weeks at 4&#xa0;℃, − 20&#xa0;℃, and − 80&#xa0;℃ recovered the highest viabilities at 4&#xa0;℃ for most cultivars, although differences in mean viability between storage treatments were not statistically significant. In addition to pollen viabilities, this study describes certain details of microgametophyte structure and development, including pollen tube development and the production of two sperm nuclei at anthesis.</p>
