In Vitro Anticancer Potential of Rauvolfia tetraphylla L. on Hepatocellular Carcinoma

Rauvolfia tetraphylla L. commonly known as, Wild Snake Root is a woody shrub in family Apocynaceae. The goal of this study was to investigate R. tetraphylla’s anticancer potential and its cellular mode of action against the HeP-G2 cell line. R. tetraphylla powdered leaf and root were extracted in 70% ethanol using the hot continuous percolation technique. The crude ethanolic leaf and root extracts were tested for MTT assay, ethidium bromide and acridine orange dual staining, changes in mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) production. Both leaf and root extracts decreased HeP-G2 cell viability, with IC50 values of 32.64 g/ml and 50.28 g/ml, respectively. Apoptosis was the major type of cell death determined by EtBr/Ao staining in Hep-G2 cells upon both treatments. Both leaf and root extract treatments for 24 h induced depolarization in MMP and production of ROS. The leaf extract showed higher cytotoxic effect on HeP-G2 cell line as compared to root extract. Collectively, our findings demonstrated that the application of plant treatment triggered the generation of ROS and caused a reduction in the MMP. These effects could potentially have contributed to the apoptosis of HeP-G2 cells.
