Effects of Salt Stress on Seed Vigor and Water Migration in Adzuki Beans

Salt stress is a key factor that affects seed vigor. To explore the effects of salt stress on the water status of adzuki bean seeds and its correlation with seed vigor, we used low-field nuclear magnetic resonance to investigate germination rate and water migration in adzuki bean seeds under different NaCl concentrations (0, 30, 60, 90, 120, and 150 mmol/L). The results indicated that with an increase in NaCl concentration, the germination potential, germination rate, and vigor of adzuki bean seeds decreased. When the NaCl concentration reached 150 mmol/L, the seed germination rate dropped to 3%, indicating that NaCl stress inhibited the germination of adzuki bean seeds to different degrees. The amounts of free and bound water both showed a decrease in trend after 24 h of stress and with increase in NaCl concentration. As the NaCl concentration increased, the proportion of free water decreased from 3.835 in the control group to 2.869 in seeds treated with 150 mmol/L NaCl. As the duration of stress increased, the proportion of free water increased. Under 60 mmol/L NaCl stress, with increase in stress duration, the free water content increased from 31.880 to 3466.395, and the bound water content increased from 488.193 to 721.008. The ratio of free water to bound water gradually increased, and the proportion changed from 0.065 to 4.808. The results of this study on the effects of salt stress on plant seed germination and changes in water content provide a new perspective for understanding the mechanisms of salt stress in plant seeds. It also offers new ideas and strategies for improving seed selection and application in saline–alkali soils.
