STIM2 suppressed Anti-tumor immunity in cholangiocarcinoma

Stromal interaction molecule 2(STIM2) has been implicated in the progression of various kinds of tumors. Existing researches have not explored its effect on how tumor-associated macrophages (TAMs) are polarized into the M2 phenotype in cholangiocarcinoma. CCA cells underwent co-culture with an M2-like conditioned medium (M2-CM). We established the subcutaneous xenograft model of nude mice. The western blot together with quantitative real-time PCR (qPCR) assisted in the determination of molecule expression. STIM2 suppresses anti-tumor immunity in CCA through inhibiting CD8 + T cell under the mediation of TAM. STIM2 induces TAMs to be transformed into pro-tumorigenic M2-like phenotypes through the activation of cAMP signaling pathway. STIM2 holds a large potential of serving for the immunotherapy targeting CCA by virtue of TAMs. The molecular mechanism regarding its tumor-promoting effect in CCA is elucidated in the study.
