Overexpression of PsLHY, a novel MYB-related gene from tree peony (Paeonia suffruticosa), enhances vegetative growth duration and boosts fruit yield in tobacco

The tree peony (Paeonia suffruticosa) is extensively cultivated due to its significant economic value in ornamental, medicinal, and oil production. MYB-related proteins, particularly LHY (Late Elongated Hypocotyl), which are part of the CCA1 (Circadian Clock Associated 1) family, play crucial roles in circadian clock function and flowering regulation. However, the evolutionary paths and functional diversification of MYB-related family members in tree peony are not yet understood. In this study, 35 MYB-related genes were identified from the transcriptome of tree peony, which were categorized into five groups based on phylogenetic analysis. Through expression pattern and correlation analysis, PsLHY, a novel MYB-related gene, was obtained. PsLHY was expressed in different tissues in tree peony, with the highest levels observed in sepals and leaves. The expression patterns of PsLHY and genes related to flowering differed before and after flowering and also changed during the day, with higher expression observed in the light than in the dark. The length of the PsLHY coding sequence (CDS) was 1,854 bp, encoding 617 amino acids with a MYB DNA-binding domain. Further transcriptional activation activity analysis revealed strong activation activity at the C-terminus of PsLHY. Overexpression of PsLHY in tobacco (Nicotiana tabacum) resulted in prolonged vegetative growth and increased fruit and seed yield, potentially through the regulation of PsFT and PsTFL1 expression. The study provides valuable insights into the functional characterization of MYB-related proteins in tree peony and offers a novel genetic resource for high-yield breeding of this species.
