Development and validation of a simple RP-HPLC method for quantification of bergenin in different parts of Bergenia ciliata (Haw.) Sternb

Bergenia ciliata (Haw.) Sternb. is an important medicinal plant found in the temperate Himalayan region. Rhizomes of Bergenia ciliata are used in traditional systems of medicine for treatment of kidney and bladder stones. Bergenin is a major bioactive compound found in the rhizomes of Bergenia ciliata, which reportedly exhibits antioxidant, anti-inflammatory and anti-microbial properties. A new RP-HPLC method has been developed for quantification of bergenin in the rhizomes and leaves of Bergenia ciliata. The method developed was found to conform to International Council of Harmonisation guidelines (ICH-Q2(R1)) and was robust, accurate and precise. The analysis was conducted using a Sunfire C-18 column (4.6 × 250 mm, 5 μm) with gradient elution, where solvent A was 0.1 per cent formic acid in water (pH = 2.5) and solvent B was acetonitrile. The detection wavelength was set at 278 nm and column temperature was maintained at 32 °C. A flow rate of 0.6 mL/min was maintained over the 15 min runtime. The method demonstrated good linearity (R² = 0.999) over a broad concentration range from 6.25 to 150 µg/mL. The relative standard deviation (RSD) was less than 2 per cent in inter-day and intra-day precision. The average recovery for bergenin ranged from 100.164 to 101.980 per cent and levels of LOD and LOQ were 0.00305 and 0.0122 µg/mL, respectively. The bergenin content was quantified in the rhizomes and leaves of Bergenia ciliata, which was 1.027 and 0.253 per cent w/w, respectively. Compared with previously reported methods, the present method offers reduced runtime and flow rate while maintaining satisfactory sensitivity, precision and accuracy. The validated method is suitable for routine quantification of bergenin and may serve as a useful tool for bergenin-based quality evaluation of Bergenia ciliata raw materials and herbal preparations.
