Background <p>Atorvastatin is a lipophilic statin that exhibits pleiotropic effects beyond cholesterol (Chol) reduction. Sortilin is one of the factors related to insulin sensitivity and Lipid metabolism. Sortilin, as a cell surface and intracellular receptor, is a main member of the vacuolar protein sorting 10 proteins (VPS10Ps) family.</p> Objective <p>This study aimed to evaluate the effects of atorvastatin consumption on blood levels of sortilin, glycemic, and Lipid indices in patients with type 2 diabetes mellitus (T2DM).</p> Methods <p>A randomized controlled trial was conducted among 60 participants, who were grouped into three distinct categories: patients with T2DM, pre-treatment with atorvastatin 40&#xa0;mg/day into group 1 (<i>n</i> = 30); patients with T2DM, after treatment with atorvastatin 40&#xa0;mg/day, group 2; and post-treatment and healthy controls, group 3. Patients used atorvastatin for 3 months.</p> Results <p>FBS, HbA1c, LDL-C, TG, T-Chol, insulin, HOMA-IR, and sortilin concentrations in group 1 were significantly higher than those in group 3. But there was no significant difference in HDL-C content between the two groups. FBS, HbA1c, HDL-C, insulin, HOMA-IR, and sortilin concentrations in the diabetic group 2 were notably&#xa0;higher than those in group 3. But the serum levels of LDL-C in group 2 receiving atorvastatin are lower than those in group 3. The two groups had no notable difference in TG and T-Chol serum levels. Atorvastatin significantly attenuated the blood levels of FBS, HbA1c, TG, T-Chol, LDL-C, HOMA-IR, and sortilin. But the serum levels of HDL-C and insulin notably increased by atorvastatin.</p> Conclusion <p>Atorvastatin can attenuate diabetic complications by improving glycemic indices, lowering TG, LDL-C, and sortilin concentrations, and enhancing HDL-C levels.</p> Trial registration <p>This randomized clinical trial was registered at IRCT.ir on 2023-12-12 (Registration Code: IRCT20231122060137N1).</p>

Background

Atorvastatin is a lipophilic statin that exhibits pleiotropic effects beyond cholesterol (Chol) reduction. Sortilin is one of the factors related to insulin sensitivity and Lipid metabolism. Sortilin, as a cell surface and intracellular receptor, is a main member of the vacuolar protein sorting 10 proteins (VPS10Ps) family.

Objective

This study aimed to evaluate the effects of atorvastatin consumption on blood levels of sortilin, glycemic, and Lipid indices in patients with type 2 diabetes mellitus (T2DM).

Methods

A randomized controlled trial was conducted among 60 participants, who were grouped into three distinct categories: patients with T2DM, pre-treatment with atorvastatin 40 mg/day into group 1 (n = 30); patients with T2DM, after treatment with atorvastatin 40 mg/day, group 2; and post-treatment and healthy controls, group 3. Patients used atorvastatin for 3 months.

Results

FBS, HbA1c, LDL-C, TG, T-Chol, insulin, HOMA-IR, and sortilin concentrations in group 1 were significantly higher than those in group 3. Atorvastatin significantly attenuated the blood levels of FBS, HbA1c, TG, T-Chol, LDL-C, HOMA-IR, and sortilin. But the serum levels of HDL-C and insulin notably increased by atorvastatin.

Conclusion

Atorvastatin can attenuate diabetic complications by improving glycemic indices, lowering TG, LDL-C, and sortilin concentrations, and enhancing HDL-C levels.
