Enzymatic hydrolysis of fermentable sugars prepared from bamboo by pretreatment with hot water and green liquor

Pretreatment is the key step in enzymatic hydrolysis of bamboo wood. Green liquor pretreatment is a promising method to improve enzymatic hydrolysis and saccharification efficiency by changing the physical and chemical properties of lignocellulose surface. At present, there is little work to quantitatively analyze the enzymatic saccharification rate of bamboo lignocellulose by two-stage pretreatment. In this study, a two-stage pretreatment method combining hot water and green liquid phase was mainly used, and the bamboo was treated with Bovine serum albumin (BSA) and Tween80 (TW) chemical additives. After that, the treated samples were hydrolyzed by cellulase, and the effects of pretreatment on the chemical composition and physical properties of bamboo and the influencing factors of chemical additives on the enzymatic hydrolysis efficiency of bamboo were explored. The results showed the two pretreatment stages of hot water and green liquid to have an evident effect on the enzymatic hydrolysis of bamboo. The two-stage pretreatment with hot water and green liquid (14% total alkali) resulted in an 85.51% yield of total sugar hydrolyzed by the enzyme. The addition of BSA and Tween80 improved the efficiency of the enzymatic hydrolysis of bamboo, and the synergistic effect of them could make the total sugar yield of enzymatic hydrolysis reach 99.67%. Through calculation, 65.88 g of fermentable sugar can be obtained per 100 g of bamboo fiber through enzymatic hydrolysis with cellulase.
