<p>A series of novel thienopyrimidine derivatives were synthesized and evaluated for their antimicrobial activity. The target compounds were obtained through a multi-step synthetic approach, including Mannich base reactions. The synthesized compounds demonstrated promising antibacterial and antifungal activities, with some compounds exhibiting MIC values as low as 3.125&#xa0;µg/mL. Molecular docking studies were conducted to investigate the binding interactions of the most active derivatives with the 1DLs protein, showing significant binding affinities comparable to standard antibiotics. These findings suggest that the synthesized compounds have potential as new antimicrobial agents, warranting further optimization and development.</p>
