Exploration of ERK/Myc regulating ribosomal biosynthesis and promoting ALPPS related liver regeneration

To research the mechanism of ribosome biosynthesis in liver regeneration after associated liver partition and portal vein ligation for staged hepatectomy (ALPPS). 50 healthy male SD rats were randomly divided into PVL group and ALPPS group. After 1, 2, 3, 5 and 7 days after operation, the liver regeneration rate was calculated. The expression of ribosomal transcription product 47 S-preRNA was detected in the right middle lobe (RML) by QRT-PCR, and the transcription rate of rDNA was detected by AgNOR staining. The expression of ribosomal biosynthesis markers c-Myc and ERK was detected in the RML by Western blot. FLR tissue from 3 patients with ALPPS was used for validation. The liver-to-body weight ratio was markedly higher in the ALPPS group on postoperative days 1, 3 and 7 than in the PVL group (P < 0.05). The number of silver-stained particles per nucleus in the RML was significantly higher in the ALPPS group on postoperative day 1. The expression level of 47 S-preRNA was stronger on the 1st and 3rd day after operation in ALPPS. The expression of ERK and c-Myc was more significant in ALPPS group at 1st, 2nd and 3rd day after operation. In addition, the expression of 47 S-preRNA, ERK and c-Myc was significantly higher in the FLR tissues of ALPPS-II stage patients compared with ALPPS-I stage. The rate of ribosome biosynthesis was significantly upregulated during ALPPS. ALPPS may accelerate ribosome biosynthesis through ERK/Myc pathway, so as to promote rapid liver regeneration.
