Recovery model of male salivary DNA from female skin: a comparative analysis

Forensic investigations surrounding sexual assaults often look for the presence of semen as major biological evidence. In most of the cases, semen and blood are not available. In such cases, saliva can be examined and used for identification of the perpetrator of the crime. Saliva is transferred from one body to another during any assault, knowingly or unintentionally. The presence of saliva on the victim’s body must be checked to help in the positive outcome of the investigation. This study simulates the saliva exchange between the male and female, similar to a transfer in any assault. The male volunteers were advised to deposit saliva onto the forearm of a female. The study compared the efficacy of three commonly used DNA extraction methods—organic, chelex, and silica column based. In our study, the silica column based method yielded more DNA than the other two; this outcome correlates with the genotyping results showing amplification at all 38 Y-STRs loci of Ingenomics Y Profiler kit, with a success rate of 83% for silica column based kit. Additionally, the study compared the two collection methods—single and double swabbing, which statistically showed no significant difference but yielded good amount of DNA.
