Elucidation of genetic diversity in the medicinal herb Tribulus terrestris L. from North Indian plains using morphological and ISSR markers

Tribulus terrestris L. of family Zygophyllaceae is a highly important medicinal herb that grows wild along the roadside and on wastelands. Field surveys were conducted at twenty-two sites of Delhi, Himachal Pradesh, Haryana, Jammu, Punjab, Uttarakhand, Uttar Pradesh and Rajasthan to study morphological diversity and collect samples for DNA analysis. Quantitative trait’s data showed that the maximum average plant length (77.33 cm) was recorded from Bulandshahr, highest average root length (17.76 cm) was observed at Sonipat and maximum average number (10.33) of flowers per plant was observed at Dausa. Genetic diversity was examined at the molecular level using Inter-Simple Sequence Repeat (ISSR) markers. In total, 16 primers produced 94 loci in a size range of 200–2000 bp. Maximum (9) fragments amplified by UBC-868. Mean Polymorphism Information Content (PIC) and mean Marker Index (MI) were 0.426 and 2.258, respectively. Dendrogram showed three major groups. Factorial analysis also placed all accessions in concordance to the groupings of the dendrogram. Bayesian analysis using STRUCTURE showed three genetic stocks for the analyzed accessions. This is the first study covering large area and diverse accessions of T. terrestris from North India. The results of present study can be useful in selection and management of elite accessions which can be used by future researchers for improvement, and herbalists for making more effective formulations.
