Establishment and Evaluation of Immunocolloidal Gold Chromatography Method in the Sandwich Mode for Novel Coronavirus Double-Antibody Detection

To facilitate the prompt diagnosis of SARS-CoV-2 infection, a sandwich immunocolloidal gold chromatography assay targeting the envelope proteins of SARS-CoV-2 was developed. BALB/c mice were immunized with purified recombinant SARS-CoV-2 E protein. Positive hybridoma cells were generated through standard fusion techniques, and the corresponding monoclonal antibodies (mAbs) were isolated from ascites fluid using the caprylic acid–ammonium sulfate precipitation method. Subsequently, a double-antibody sandwich immunocolloidal gold chromatography system was constructed for diagnostic application. The sensitivity, specificity, and concordance rate of the methodology were rigorously assessed.
