Optimization of microwave extraction and antioxidant activity of swertisin from Thlaspi arvense seeds using high performance liquid chromatography detection and response surface methodology

To optimize the microwave extraction process of swertisin from Thlaspi arvense seeds, this paper first establishes an HPLC detection method for swertisin in T. aivense. Based on the single factor experiment, Design-Expert 11 software and Box-Behnken Response Surface Methodology (RSM) were used based on the single factor experiment to optimize the microwave extraction process with swertisin content as an index. Additionally, the antioxidant activity of the ethanolic extract of T. arvense seeds was tested. The optimum response surface optimization conditions for microwave extraction were as follows: microwave power 420 W, extraction time 156 s, a solid-liquid ratio of 14:1 (mL/g), and an ethanol concentration of 54%. Under these conditions, the content of local swertisin in T. arvense seeds was 0.518 ± 0.061 mg/g. Scanning electron microscopy revealed that the degree to which the cell structure of the residues was destroyed after extraction was indicative of the amount of swertisin obtained by different extraction methods. The ethanol extract of T. arvense seeds demonstrated DPPH and OH radical scavenging activities with IC50 values of 52.273 mg/mL and 103.550 mg/mL, respectively. Moreover, the extract exhibited a progressive increase in Fe3+ reducing activity as concentrations rose from 0.2 to 1.0 mg/mL, that ethanol extract of T. arvense seeds has good antioxidant activity. The HPLC method established in this paper was simple, the optimized extraction process of T. arvense was stable, the antioxidant activity of the ethanol extract was good and can be used for the extraction of swertisin from seeds of T. arvense.
