<p>The present work aimed to determine the toxicity effect of microencapsulated <i>Lactobacillus plantarum</i> RS09, previously isolated from fermented finger millet porridge and incorporated in millet-based probiotic chocolate. The probiotic millet-based chocolate was assessed for its safety using acute and subacute oral toxicity tests on Wistar albino rats. No mortality or treatment-related illness was observed regarding body weight, behaviour, water intake or feed consumption during the acute oral toxicity study. Results showed that the microencapsulated <i>L. plantarum-</i>RS09 probiotic millet chocolate was safe up to 3000&#xa0;mg/kg in a 14-day treatment period. Both acute and subacute oral toxicity studies showed neither toxicity effects nor death in rats. The samples were administered at two dosage levels: low dosage (300&#xa0;mg/kg) and high dosage (900&#xa0;mg/kg). No significant difference in haematology and biochemical parameters was observed between the control and treated groups. Histopathology studies of liver, spleen, kidney and ileum exhibited no abnormalities. The RS09 strain was also able to reduce harmful intestinal pathogenic bacterial populations and increase the lactic acid bacterial population. Thus, microencapsulated <i>L. plantarum</i> RS09 millet-based chocolate is safe, non-toxic and could be used as a potential probiotic for consumption.</p>
