Zeatin-induced micropropagation of potato cv. Pilak local and its fidelity assessment

Potato (Solanum tuberosum L.) (Solanaceae), an herbaceous temperate crop, is monitored as a critical food security crop because it is consumed as an alternative to many cereals in low-resource areas. In addition to the popular genotypes, indigenous genotypes of potato, like cv. Pilak local, are highly rich in nutrients and have an untapped source of wild genes; nonetheless, these are cultivated in patches only in northeastern India. There is a need for interventions to bring this cultivar into mainstream cultivation. Therefore, cultivar-specific protocols for micropropagation and assessment of genetic fidelity were established for the first time in Pilak local. Fresh sprouts were inoculated in Murashige and Skoog (MS) media supplemented with 6-benzyladenine, kinetin, meta-Topolin, and zeatin (Zea) at 0.5–1.5 mg/l. The maximum shoot proliferation was recorded in the 1.5 mg/l Zea treatment. The shoots were rooted in MS media supplemented with 1.0–3.0 mg/l indole-3-acetic acid or indole-3-butyric acid (IBA), and the maximum rooting was recorded at 1.0 mg/l IBA. Monophasic multiple shoot and root formation was achieved successfully in MS media with combinations of 1.0 mg/l IBA with variable Zea levels. The combination of cytokinin and auxin (1.5 mg/l Zea + 1.0 mg/l IBA) had the best results. The micropropagated plantlets were hardened in cocopeat with 95% survival and then grown in the field. During genetic fidelity analysis, analogous peaks of DNA in the flow cytometry system and monomorphic banding patterns in the four molecular marker systems confirmed the true-to-type nature of the plantlets. The present protocol would be of high commercial importance, as this is the first report on this cultivar.
