In Vivo Synergistic Effect of Polyherbal Extract, Design and Development of a Mouth-Dissolving Tablet for Diabetes Mellitus Management

This study formulated a mouth dissolving tablet containing Emblica officinalis (Amala), Terminalia chebula (Hirda) and Glycyrrhiza glabra (Liquorice) extract for Diabetes Mellitus treatment. The synergistic antidiabetic effect of the polyherbal aqueous extract were evaluated using the OGTT on STZ- induced wistar rats over 21 days. The MDTs were prepared using Direct Compression Method, and their physical appearance, weight uniformity, hardness, friability, wetting time, disintegration time, dissolution, and stability were assessed. After 21 days, blood sugar levels dropped to 150–170 mg dL− 1 due to considerable reduction caused by the polyherbal extract (50 mg of each herb), while metformin kept blood sugar levels at normal levels (70–100 mg dL− 1). The optimised formulation F6 batch had a minimal disintegration time of 1.16 ± 0.155 min and 99.02% drug release. Blood glucose level was significantly lowered by the polyherbal extract in diabetic rats compared to diabetic control rats with STZ induced diabetes. But it was not as potent as metformin in lowering blood glucose. The optimized formulation for tablet containing mouth dissolving matrix produced satisfactory pharmaceutical properties, such as high in vitro release of drug and rapid disintegrating property.
