pH-Sensitive Peptide-Modified Nanoliposome Containing Berberine for Prostate Cells (DU145) Targeting and Anti-tumor Activity

Modern cancer treatment relies on a balance between targeted and non-targeted toxic effects. The acidic environment of tumors is a target for therapy pH-sensitive liposome deliver drugs upon encountering this acidity. Berberine (Ber), a natural compound with anticancer properties, is investigated here for targeted delivery using liposomes in prostate cancer treatment. In this study, the anticancer effect of pH-sensitive nanoliposomes, surface-modified with RGD peptide and containing Ber, was evaluated in the DU-145 cell line expressing αVβ3 integrin. The results showed that the highest amount of drug released from pH-sensitive nanoliposome is in pH 6.5. MTT assay revealed significant toxicity of pH-sensitive liposomes compared conventional liposomes in pH 6.5 medium. Moreover, the apoptosis assay using annexin V/propidium iodide confirmed cytotoxicity data for significant efficacy of pH-sensitive liposomes. Ultimately, these results revealed high potential of pH-sensitive peptide-modified nanoliposome containing Ber for inhibiting the proliferation of DU-145 cells compared to the non-targeting nanoparticles.
