<p>A crude extract rich in sulfated polysaccharides (SPL), from <i>Pistacia lentiscus</i> L. leaves was comprehensively investigated to determine its chemical composition, water-holding capacity, bioactivity, and gastroprotective effects in an ethanol-induced gastric ulcer model. SPL exhibited high levels of uronic acids (434.52 ± 3.68 mg GlcAE/g) and sulphate groups (603.96 ± 0.51 mg K₂SO₄E/g). Its water-holding capacity, recorded at 4 ± 0.1 g water/g SPL, reflects promising functionality for biomedical or nutraceutical applications. Furthermore, the SPL exhibited substantial antioxidant activity, with IC₅₀ values of 17.65 ± 0.63 µg/mL in the β-carotene bleaching assay and 73.12 ± 0.22 µg/mL for hydroxyl radical scavenging. It also showed significant urease inhibition (IC₅₀ = 39.59 ± 1.44 µg/mL) and proton pump inhibition (IC₅₀ = 0.92 ± 0.021 mg/mL), supporting its potential use in managing gastric hyperacidity and Helicobacter pylori-associated complications. In an ethanol-induced ulcer model, SPL dose-dependently reduced ulcer severity, achieving 67.52% inhibition at 200 mg/kg, along with reduced oxidative stress and enhanced catalase activity. Furthermore, SPL exhibited antibacterial activity, particularly against Staphylococcus aureus and Bacillus subtilis, with inhibition zones of 14.00 ± 1.41 mm and 11.67 ± 2.08 mm, respectively. Collectively, these findings highlight the multifunctional bioactivity of SPL, supporting its potential as a natural therapeutic agent through the alleviation of gastric tissue injuries, oxidative damage, and microbial infections.</p> Graphical Abstract <p></p>
