Purpose <p>Given the growing interest in sustainable alternatives for parasite control in livestock, in this study, the nematocidal activity of two <i>Pleurotus djamor</i> spent substrate mixtures was evaluated and purified to identify the compounds responsible for the activity.</p> Methods <p>Two separate substrate mixtures were prepared with input ratios of 0.5:0.5:2 and 1:1:1 with corn stover, Pangola grass and coffee pulp, then basidiomes of <i>P. djamor</i> were produced. Subsequently, the spent substrate from each treatment was exposed to sequential extraction with hexane, ethyl acetate, butanol and water. The in vitro nematocidal activity of the different extracts was evaluated against <i>Haemonchus contortus</i>, and the most active fractions were purified by open column chromatography. Finally, the most active fractions were analyzed by gas chromatography-coupled mass spectrometry (GC–MS) to identify the active compounds. The same was done but using substrate without mushroom as a control.</p> Results <p>The ethyl acetate extracts obtained from treatment 2 showed higher nematocidal activity, both on the non-inoculated and spent substrate. GC–MS analysis allowed the identification of “caffeine” as one of the major compounds.</p> Conclusion <p>It was determined that the amount of coffee pulp used in the mixture is proportional to the nematocidal activity observed. No difference was observed in the nematocidal activity when using substrate without inoculum with respect to the spent substrate. “Caffeine” could be responsible for the nematocidal activity.</p> Graphical Abstract <p></p>
