<p>Extract from empty pods of <i>Pisum sativum</i> (garden pea), lignocellulosic agricultural residue, was used as a substrate for <i>Bacillus subtilis</i> (a probiotic) to produce the alkaline protease enzyme. The extract was utilized as a nutrient source for submerged state fermentation. Initially, the extraction from empty pea pods was done through batch extraction, where solid loading (% w/v) and temperature varied at 350 RPM, as well as autoclave-assisted extraction, where autoclave extraction time was varied at known temperature (121°C) and pressure (15 PSI). Submerged fermentation was carried out at pH 8 with 1% (v/v) inoculum (titer of 1.5-2 × 10 cfu/mL) and incubated at a temperature of 35&#xa0;°C and an agitation speed of 130 RPM using different extracts from biomass as a substrate. In the preliminary fermentation studies, the maximum alkaline protease activity was observed when autoclave-assisted extract was used as a substrate. After further optimization, the maximum enzyme activity of 11.82 U/mL was obtained with 2% inoculum size (v/v), pH 9, temperature 35&#xa0;°C, and agitation speed 130 RPM after 6&#xa0;h using autoclave-assisted extract of 15 % solid loading and 20 minutes extraction time.</p>
