Purification and Analysis of R-Phycoerythrin from Hypnea musiformis: Insights into Structural Integrity and Purity

This study focuses on the extraction, purification and characterization of R-Phycoerythrin (R-PE) from Hypnea musciformis, a red seaweed species known for its high phycobiliprotein content. R-PE was extracted using a precipitation method with 55% ammonium sulfate, followed by purification through Q-Sepharose column chromatography, resulting in a maximum purity index 4.9. Spectral analyses revealed a fluorescence emission peak at 578 nm and absorption peaks at 498 nm, 536 nm, and 566 nm, characteristic of R-PE. Native- PAGE showed a uniform protein band at 240 KDa, confirming structural integrity post-purification. SDS-PAGE and 2D-Gel electrophoresis identified subunits (α, β, γ) and their molecular masses. These findings highlight R-PE’s robust purity and structural uniformity from Hypnea musciformis, making it suitable for potential applications in biotechnological and biomedical fields.
