Optimization of Extraction Conditions and Characterization of Lipases from Pequi (Caryocar Brasiliense Camb.) Fruit By-Product

This study evaluated the optimization of solid-liquid extraction of lipase from pequi shell using response surface methodology (RSM) as a statistical tool to determine the best conditions (time and extraction solution pH) for obtaining an extract with higher lipolytic activity (U/mg). A significant effect of extraction time and pH of the medium on the response variable was observed, with a maximum activity of ≈ 189.55 U/mg under the optimized conditions. The extract was characterized for maximum efficiency of hydrolysis reaction (EHRmax) and stability as a function of pH (4–10) and temperature (30–80 °C). EHRmax (3911.18 ± 28.28 U/mg) was observed at pH 5 and 60 °C, and the enzyme stability (relative activity ≥ 60%) was observed in the pH 5–7 range up to 180 min. Therefore, the experimental design used for the extraction was efficient, and the model found by the response surface represented the empirical values.
