Yet, ongoing efforts are crucial to identify new drug targets and develop combination therapies to address evolving tumor adaptations in these patients.</p> Recent Findings <p>In the past decade, disrupted epigenetic regulation has emerged as a critical factor in various cancers, including breast cancer. Chromatin modifiers and transcription factors play pivotal roles in disease development and treatment outcomes. Integrating novel epigenetic-based therapies with existing targeted treatments presents a promising strategy against resistance.</p> Summary <p>This review examines cutting-edge advancements in endocrine therapy and PI3K/AKT inhibitors in ER + breast cancer. Additionally, it explores pivotal chromatin-based mechanisms influencing breast cancer progression and treatment response. Lastly, it highlights the potential of targeting epigenetic regulators as a promising approach to combat therapy resistance in breast cancer.</p>
