<p><i>Diaphania indica</i> (Saunders) (Lepidoptera: Crambidae) is an important pest of snake gourd, <i>Trichosanthes anguina</i> (L.). A comparative analysis of Chlorophyll <i>a</i> and <i>b</i>, total chlorophyll, proteins, phenols, reactive oxygen species (ROS), hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>), malondialdehyde (MDA) and antioxidative enzymes [catalase (CAT), superoxide dismutase (SOD), glutathione S-transferase (GST), ascorbate peroxidase (APOX), pyrogallol peroxidase (PPX) and guaiacol peroxidase (GPX)] of three snake gourd cultivars, MNSR-1 (MNS), Baruipur Long (BAR) and Polo No. 1 (POLO) after 4 or 12&#xa0;h of <i>D. indica</i> feeding were conducted to observe the induced defense mechanism of these cultivars (<i>n</i> = 5). 3,3ʹ-Diaminobenzidine (DAB) and nitro blue tetrazolium (NBT) staining, and expression of <i>TaCAT3</i> gene were observed in undamaged and insect-damaged cultivars (<i>n</i> = 3). Chlorophyll <i>a</i> and <i>b</i>, total chlorophyll and proteins decreased with increased feeding time in each cultivar. Total ROS, H<sub>2</sub>O<sub>2</sub>, MDA, phenols and antioxidative enzymes were enhanced with increased feeding time in each cultivar. After 12&#xa0;h of feeding, DAB staining was 5.43 and 8.71% higher in POLO than MNS and BAR (<i>P</i> &lt; 0.05), respectively, whereas NBT staining was 8.68 and 30.22% higher in POLO than MNS and BAR (<i>P</i> &lt; 0.05), respectively. In contrast, significant increases in activities of enzymatic antioxidants (CAT, PPX and GPX) and total phenols were observed in POLO than MNS and BAR (<i>P</i> &lt; 0.05). After 12&#xa0;h of insect feeding, induction of <i>TaCAT3</i> gene was significantly higher in POLO than MNS (<i>P</i> &lt; 0.05). This study concludes that MNS and BAR were more susceptible to <i>D. indica</i> than POLO.</p>
