<p>Two genes encoding myofibril-bound serine protease (MBSP; LOC109099648 and LOC109072694) are aligned in tandem between <i>gnb5b</i> and <i>myo5a</i> on common carp chromosome B18. Chromosome 18 of cypriniform fish carries an orthologous gene, but this gene is absent in the genomes of other Ostariophysi fish. Numerous genes encoding trypsin-like serine proteases are aligned with the trypsin gene in Ostariophysi fish genomes. Cypriniform MBSP shares a conserved region with some trypsin-like serine proteases, and these serine proteases form a single cluster in the phylogenetic tree. LOC109099648, trypsin (<i>prss1</i>), and trypsin-like seine protease genes (LOC109097175) were expressed in the ordinary muscle and hepatopancreas of common carp, whereas LOC109072694 was expressed only in ordinary muscle. Amino acid sequence predictions showed a glutamic acid at the C-terminus of the signal peptide of common carp and goldfish MBSP, whereas Ostariophysi trypsin and trypsin-like serine proteases have a lysine at the same position. These results suggest that the MBSP gene might have emerged via duplication of a common ancestor to the trypsin gene, followed by transposition to chromosome 18 after an ancestor of cypriniform fish had separated from other Ostariophysi fishes. Common carp and goldfish MBSP may be further evolving to exhibit unique control of gene expression and activation.</p>
