Changes in sperm quality and seminal plasma parameters of masu salmon Oncorhynchus masou during the breeding season and implications for cryopreservation success

Effective cryopreservation of fish sperm is vital for sustainable aquaculture and conservation. In masu salmon Oncorhynchus masou, milt quality varies throughout the breeding season, influencing artificial breeding and cryopreservation success. This study examined seasonal variations in sperm motility and seminal plasma properties to determine optimal milt collection timing and key predictors of post-thaw viability. Milt from six Mori-strain males was collected at four intervals (mid-September, late September, mid-October, and late October), totaling 24 males. Samples were analyzed for motility, viability, and concentration before and after freezing. Computer-assisted sperm analysis was used to assess motility, while seminal plasma pH, electroconductivity, DNA concentration, total protein concentration, and osmolality were also measured. Significant changes in motility were observed, with peak performance in late September. Notably, samples with similar initial motility often exhibited variable post-thaw outcomes, suggesting that initial motility alone does not predict cryopreservation success. Seminal plasma pH and electroconductivity correlated positively with post-thaw motility, whereas DNA and protein concentrations showed negative correlations. Based on these findings, we recommend freezing milt with high initial motility, seminal plasma pH around 8.3, and electroconductivity between 11.7 and 18.2 mS/cm. These results underscore the importance of seminal plasma assessments alongside motility data to improve cryopreservation outcomes in O. masou.
