<p>Increased environmental pollution has a deleterious influence on sperm parameters and sexual health among individuals exposed to air pollution. Cadmium (Cd) and lead (Pb) are heavy metals present in the environment that disrupt the endocrine system, leading to decreased spermatogenesis and sperm quality in males occupationally exposed to air pollution. The current research work aims to investigate the primary sources of heavy metals (Cd and Pb) and their effects on amino acids in the seminal plasma of normal and infertile individuals. Ultra-Performance Liquid Chromatography (UPLC) was used to quantify critical amino acids in the seminal plasma of normal and infertile groups. Sperm viability and hypo-osmotic swelling (HOS) testing were performed to assess sperm activity and function. Cd and Pb may inhibit enzymes such as dihydrolipoamide dehydrogenase (DLD), a component of the branched-chain α-keto acid dehydrogenase complex. This results in poor branched-chain amino acid (BCAA) catabolism and reduced substrate availability for the tricarboxylic acid (TCA) cycle, leading to decreased ATP synthesis essential for spermatogenesis. Isoleucine, leucine, and valine are critical amino acids found in human seminal plasma and are required for mitochondrial activity, with the sperm-rich fraction containing the highest quantities. Statistical analysis showed a significant difference in amino acid levels between the normal and infertile groups (<i>p</i>-value &lt; 0.05). Our findings highlight the impact of heavy metals Cd and Pb on seminal plasma amino acid composition and mitochondrial activity, ultimately compromising sperm quality in exposed individuals.</p>
