Blood Ten Eleven Translocase 3 Methylation Levels as a Marker to Assess Response to Treatment in Head and Neck Carcinoma Patients

Ten-eleven translocase (TET) enzymes are responsible for DNA demethylation reactions and play a key role in regulating the methylation balance in the body. TET expression instability may result in methylation alterations. We performed methylation specific-polymerase chain reaction to evaluate the TET3 promoter hypermethylation in the DNA isolated from the venous blood samples of head and carcinoma (H&NC) cases before initiation of treatment (n = 32) and after treatment (n = 30) and healthy controls (n = 32) in this study. TET3 promoter was found to be hypermethylated in H&NC patients before initiation of treatment compared to after-treatment and healthy controls. No significant difference was observed between after-treatment patients and healthy controls. TET3 gene promoter methylation was significantly correlated with 5-hydroxymethylcytosine levels (5-hmC) in H&NC patients. TET3 methylation can be used as a biomarker to assess response to treatment in H&NC patients.
