Literature Review and Case Summary: Exploring Deafness as an Adverse Reaction to Bortezomib

Bortezomib has been widely utilized in the treatment of hematological system diseases including multiple myeloma (MM), Waldenstrom macroglobulinemia (WM), etc. As the first generation proteasome inhibitor approved by the U.S. Food and Drug Administration (FDA), bortezomib-based therapies have changed the treatment landscape of MM at all stages and its overall survival rate has been improved remarkable. However, bortezomib frequently generates dose-limiting peripheral neuropathy (PN), which usually leads to its dose reduction or discontinuation. Besides, other side effects such as constipation, anemia, and rash occur frequently. While some rare side effects have been occasionally reported, such as hearing loss. Compared with PN, auditory side effect caused by bortezomib was rarely reported, only with few cases, which were summarized in this paper. Its mechanism, clinical characteristic, management, and prognosis were poorly described, which were discussed in this mini-review.
