Poxviruses transcribe their genes in temporal order in the host cell cytoplasm, almost entirely by self-encoded factors. These events are mainly studied in Vaccinia, a relatively harmless poxvirus with strong similarity to pathogenic subtypes. Poxviral transcription is catalyzed by a multi-subunit viral RNA polymerase, recruited to each promoter type by viral transcription factors. Cryo-EM analyses of initiation complexes revealed unique mechanisms and also analogies to eukaryotic transcription.
