Nipah virus (NiV) is a non-segmented, negative-strand RNA virus (nsNSV), characterized by a high case fatality rate and pandemic potential with no specific treatments available. Its RNA-dependent RNA polymerase (RdRp) consists of the viral L and P proteins and is responsible for both viral genome replication and transcription. We have recently determined cryo-EM structures of the NiV RdRp in both its apo and an actively elongating state. The latter represents the first such structure reported for any nsNSV and provides mechanistic insights into RNA synthesis, which could potentially guide the development of antiviral drugs.
