The lateral organization of voltage-dependent calcium channels in lipid membranes is crucial for their function. To visualize and quantify their nanoscale arrangement, we labeled calcium channels via the HaloTag strategy and imaged them with STED microscopy in live cells and supported plasma membranes derived from giant plasma membrane vesicles. The latter further improved lateral resolution and allowed quantification of protein density and the number of channels within a cluster.
