<p>Iron-sulfur (FeS) clusters require a complex biosynthesis pathway, despite the simplicity of these versatile, inorganic protein cofactors. The process starts in mitochondria by action of the iron-sulfur cluster assembly (ISC) machinery and is the essential and minimal function of these organelles. The ISC system further assists the cytosolic iron-sulfur protein assembly (CIA) system in maturing FeS proteins in both cytosol and nucleus. Here, I briefly summarize our current mechanistic knowledge of this essential process of life.</p>
