Enhanced production of biotin in Pseudomonas mutabilis with systematic culture optimization and fed-batch strategies

Biotin has wide applications in healthcare, cosmetics, and food supplements, and traditionally is produced by chemical methods. However, an increasing attention has been shifted toward environmental-friendly biotechnological production. In this study, to improve biotin production with our constructed Pseudomonas mutabilis ZJU 3, the effects of medium components and culture conditions were systematically investigated the shake flasks, and the highest biotin (54.3 mg/L) was attained by using the optimal medium and culture conditions. Further studies showed that glycerol feeding could enhance biotin significantly, and the control of relatively lower dissolved oxygen (around 5%) improved the productivity of biotin to 351.8 mg/L in 5 L fermenter after 174 h’ cultivation. This work is very helpful to further large-scale production of biotin in the future.
