Technology Trajectories and Technology Fusion in the Glucagon-Like Peptide-1 (GLP-1) Drug Field: A Method of Main Path Analysis and Topic Modeling Based on Patent Data

GLP-1RA drugs are gaining attention in the field of diabetes due to their high efficacy, low risk and ability to treat diabetic complications. Currently, there is no study using patent analysis methodology in the field of GLP-1 drugs. The aim of this study is to identify the trajectory of technology development and technology fusion in the field of GLP-1 drugs through patent analysis.
MethodsIn this study, patent data from 1984 to 2024 were collected using the incoPat Patent Data Retrieval Library, and clustering analysis was performed using Louvain’s algorithm, primary path analysis using Pajek software, and topic identification using LDA topic model.
ResultsLong-term, oral, multi-target innovation and indication expansion of GLP-1 drugs are still the main scientific objectives nowadays. The research on albumin fusion, DPP-4 resistant GLP-1 peptide, and oral administration have shown different development directions. In addition, Pulmonary delivery formulations of GLP-1 technology and non-peptide heterocyclic GLP-1RA technology may be hot spots for future research. The infusion of medicinal chemistry technologies affects each stage of the GLP-1 drug field. Six key technical points, PTP1B inhibitors, SGLT-2 inhibitors, autoimmune diabetes, preparation of GLP-1 receptor agonist analogues, multi-receptor agonists/antagonists, and genetic engineering production, are fusion into different technological phases due to their unique technical characteristics.
ConclusionThis study conducted research on the patent data in the GLP-1 drug field. Based on clarifying the core development direction of the technology, it identified three branches in the R&D strategies of the technology and two unique technology clusters. Five technology topics and six unique technology points that are infused into the GLP-1 drug field were identified. This study facilitates researchers to quickly understand the overall development of the field, capture the key points of technology development, and discover possible future technological breakthroughs.
