Induction of Tolerance to Cryogenic Stress in Solanum tuberosum by Hydrogen Peroxide Mediated by Enzymatic Antioxidant Activity

Cryopreservation allows preservation of germplasm for long periods. Cryogenics involves oxidative stress. Hydrogen peroxide as a signal, is involved in tolerance to stress factors. The objective was to evaluate the effect of hydrogen peroxide on the induction of tolerance to cryogenic stress and the relationship with antioxidant enzymes. Pre-treatments with hydrogen peroxide for one hour one month before cryogenics, induced significant tolerance to cryogeny. Tolerance was associated with changes in the enzymatic antioxidants catalase and peroxidase. Induction of tolerance could be mediated by an increase in hydrogen peroxide internal concentration before cryogenics, suggesting the signaling role of this molecule in the long term. This work demonstrates the potential use of hydrogen peroxide in the culture phase prior to cryogenics, inducing long-term effects on enhancement of survival, reduction in catalase activity, increase in hydrogen peroxide content, and a higher water potential before cryogenics.
