<p><i>Lycium barbarum</i> L, the dry and ripe fruit of the <i>Lycium</i> genus in the Solanaceae family, is a medicinal and food homologous herb, mainly distributed in northwest of China. However, with the expansion of market demand and increasing output, the phenomenon of <i>Lycium barbarum</i> adulteration is also getting worse, so the quality control requirements of <i>Lycium barbarum</i> are becoming more and more strict. A fast and efficient quality evaluation technique is significant and necessary. In this paper, we innovated a new quality control method for <i>Lycium barbarum</i> by using a combination of one-dimensional (1D) and two-dimensional (2D) quantitative nuclear magnetic resonance (NMR) technology. And it is found that <i>Lycium barbarum</i> mainly contains glucose and fructose in nearly equal proportions. In order to achieve accurate quantification, the experimental methods and parameters are optimized and validated. In addition, we simulated the interaction between glucose, fructose, and TLR4/MD2 using Discovery Studio software, to provide some reference for studying the immunoregulatory effects of <i>Lycium barbarum</i>. In short, using the NMR technique to evaluate the quality of <i>Lycium barbarum</i> is accurate, rapid, reliable, and has certain practical significance. It is expected to be a feasible and practical detection technology for the quality control of <i>Lycium barbarum.</i></p>
