Evaluation of Regulatory T Cells and their Phenotypic Subsets in Patients with Oral Squamous Cell Carcinoma of Western Rajasthan

Head and neck cancer is a significant global health burden particularly oral squamous cell carcinoma(OSCC). The present study aimed to detect some immunoregulatory changes in regulatory T cells(Tregs) frequency and their phenotypic subsets in blood by flow cytometry. This prospective study enrolled 43 cases of biopsy-proven untreated cases of OSCC attending the outpatient department of Otorhinolaryngology of our Institute. Fifteen healthy age and sex-matched volunteers served as normal controls. Peripheral blood samples of cases and controls were processed using antibodies against CD45, CD3, CD4, CD8, CD25, CD127, and FoxP3. Analysis and gating was done to characterize subsets of Tregs on the Beckman Coulter Navious analyzer. The age of the patients ranged from 33 to 89 years with a mean age of 56.44(2SD ± 14.29) years. Among 43 cases 37(86.0%) were males and 6(14%) were females. Three Treg subsets (CD4 + CD25, CD4 + CD25 + FoxP3, and CD4 + CD25 + CD127low) were analyzed and these were increased in all the patients compared to healthy controls. Further, these were higher in cases with advanced stages of the disease. Present study emphasizes the role of regulatory-T cells in OSCC with a relatively high proportion in advanced stages. Characterization of the Tregs-subpopulation phenotype could form a strong base in the development of immunological targets for the therapeutic management of cancer patients in the future.
