Introduction <p>Meniere’s disease (MD) is an inner ear disorder characterized by episodic vertigo, fluctuating hearing loss, tinnitus, and aural fullness, with endolymphatic hydrops as the underlying pathology. While glycerol testing with pure-tone audiometry is commonly used, it remains subjective. However, Otoacoustic emissions (OAEs), including TEOAEs and DPOAEs, provide an objective measure of outer hair cell function. Despite global interest, there is limited Indian research evaluating glycerol-induced OAE changes in MD.</p> Aim <p>To investigate the frequency-specific effects of oral glycerol administration on TEOAE and DPOAE responses in individuals with unilateral Meniere’s disease.</p> Methods <p>Thirty subjects (mean age: 47.7 years) diagnosed with unilateral MD were enrolled. Pre-glycerol DPOAE and TEOAE responses were recorded at 1,000, 2,000, 3,000, and 4,000&#xa0;Hz. Each subject received an oral dose of glycerol (1.5&#xa0;ml/kg body weight) dissolved in equal saline volume. Post-glycerol OAEs were recorded after two hours using standard clinical parameters. Paired t-tests were applied to assess pre- and post-glycerol differences.</p> Results <p>Significant increases in TEOAE and DPOAE amplitudes were observed post-glycerol across all tested frequencies (<i>p</i> &lt; 0.001). The most notable amplitude enhancements occurred between 1,000 and 4,000&#xa0;Hz, suggesting improvement in outer hair cell function and inner ear fluid dynamics. These findings support the hypothesis that glycerol induces endolymph absorption, likely through osmotic effects and stria vascularis ion pump activation.</p> Conclusion <p>Oral glycerol significantly improves otoacoustic emission responses in patients with Meniere’s disease. The study highlights the diagnostic utility of frequency-specific TEOAE and DPOAE as objective tools in detecting endolymphatic hydrops.</p>
