<p>Tympanoplasty is the mainstay of treatment for patients suffering from Chronic otitis media (COM) with tympanic membrane (TM) perforation. Platelet rich plasma (PRP) is a novel biotechnology in cellular therapy which has the potential to aid in efficient TM regeneration owing to the various growth factors contained in it. This study was conducted to compare the efficacy of using PRP with respect to graft uptake and improvement of hearing, in patients undergoing Type-1 Tympanoplasty with Temporalis fascia (TF) graft. A hospital based prospective, observational, comparative study was conducted over 12 months on 120 patients aged between 18 and 45 years, suffering from mucosal type of COM. Patients were divided into 2 comparable groups matched by age, gender, laterality of perforation and surgeon expertise. Data collection was done by means of otoscopy and Pure Tone Audiometry on 30th, 60th &amp; 90th post-operative days. Our study shows a significantly higher rate of graft uptake of 97% among patients receiving PRP, as compared to 80% in the other group (p-value 0.00446). This improvement was evident across all age ranges and sizes of perforation in patients receiving PRP. The difference in mean hearing threshold gain of 18.62 dB in the PRP group and 13.13 dB in the other group was also statistically significant (p-value &lt; 0.001), proving PRP to have a significant improvement in hearing outcome. In tympanoplasty, the use of PRP along with Temporalis fascia graft helps in better graft survival and total as well as rate of post-operative hearing improvement.</p>
