<p>TWIST1, one of the primarily responsible oncogenes for epithelial–mesenchymal transition, is involved in various cancers and plays a role in metastasis. Determining the expression of TWIST1 in oral squamous cell carcinoma (OSCC) will be helpful for predicting disease prognosis and exploiting this protein as an important therapeutic target. Immunohistochemical analysis compared TWIST1 expression in normal mucosa (<i>n</i> = 30) and OSCC tissues (<i>n</i> = 60), further categorized into well-differentiated squamous cell carcinoma (WDSCC), moderately-differentiated squamous cell carcinoma (MDSCC), and poorly-differentiated squamous cell carcinoma (PDSCC). In the normal group, 13 (43.3%) samples had low TWIST1 staining, while WDSCC, MDSCC, and PDSCC showed high staining in 60%, 65%, and 90% of cases, respectively. A significant difference in TWIST1 staining index between normal and OSCC groups (<i>p</i> &lt; 0.01) was observed, with PDSCC exhibiting the highest staining. A strong TWIST1 expression was detected in 95% samples of OSCC, with being highest in PDSCC. This suggests that the expression of TWIST1 may play a role in early stages of carcinogenesis.</p>
