Immunohistochemical Expression of SOX2, Nanog, OCT4 and MENA in Salivary Mucoepidermoid Carcinoma with Relevance to Molecular Profiling

This study investigates the immunohistochemical and molecular patterns of putative stem cell transcription factors Nanog, SOX2, POU2F3, and the actin-regulatory protein MENA in forty MAML2-rearranged salivary gland MECs, with particular emphasis on delineating their contribution to tumoral molecular architecture in the context of stemness paradigms. Additionally, the POU5F1 FISH probe was used to confirm the immunohistochemical findings for OCT4. No detectable levels of Nanog, SOX2, or OCT4 were observed in any of the samples, suggesting these stem cell markers do not contribute to the development of salivary MEC. However, MENA was widely expressed in all cases. The results of the POU5F1 FISH probe were consistent with the immunohistochemical data, showing no genetic alterations in OCT4 across all forty samples. These results definitively establish that cancer stem cell populations, as defined by classical pluripotency markers, are not fundamental to the pathogenesis of salivary MEC. The pervasive expression of MENA, however, suggests alternative functional roles independent of stemness maintenance or pluripotency regulation in this particular neoplastic context.
