Modulation of Sirtuin-1 by Resveratrol as a Potential Therapy in Alleviating Metabolic Syndrome

Metabolic syndrome (MetS) is a cluster of interconnected metabolic abnormalities that significantly increase the risk of cardiovascular disease, type 2 diabetes, and other chronic health conditions. The key factors underlying metabolic syndrome include impaired glucose metabolism and dysregulated lipid metabolism, which have led researchers to explore potential therapeutic interventions. In this mini review, we aim to highlight updated findings on the therapeutic effect of resveratrol through sirtuin-1 (SIRT1) in alleviating MetS. Trans-resveratrol, the biologically active form of resveratrol, is a natural polyphenolic compound found in various fruits and vegetables. It has gained considerable attention for its potential role in ameliorating MetS through its interaction with SIRT1, a class III histone deacetylase. SIRT1 regulates cellular metabolism, aging, and several signaling pathways associated with the pathogenesis of MetS. Resveratrol-induced activation of SIRT1 boosts insulin sensitivity and promotes mitochondrial biogenesis, both of which have profound implications in managing MetS. This is supported by several studies reporting the protective effects of resveratrol against obesity, dyslipidemia, and hypertension, which are the key components of MetS. The resveratrol-induced activation of SIRT1 affects MetS through modulation of various molecular targets including peroxisome proliferator-activated receptor gamma coactivator-1 alpha, forkhead box protein O1, and adenosine monophosphate-activated protein kinase. In conclusion, the beneficial effects of resveratrol through SIRT1 activation help reverse the molecular events underlying MetS, underscoring its potential as a therapeutic agent.
