Cytotoxicity and Antiplatelet Activity of Cationic Antimicrobial Peptides of the Medicinal Leech Hirudo Medicinalis

Cationic antimicrobial peptides (CAMPs) are known as promising candidates for the development of antimicrobial drugs that do not induce resistance in pathogenic bacteria. The use of such drugs in systemic administration is the most promising hence the need to investigate the effects of CAMPs on human blood cells. Platelets are fragments of blood cells capable of forming clots to cover sites of vascular injury. In this work, the toxicity of 9 CAMPs from the medicinal leech Hirudo medicinalis to platelets was compared in the presence of blood plasma and in plasma-free media. The former conditions resulted in reduced toxicity, likely due to the binding of CAMPs to plasma components, in particular to human albumin. It was shown that all peptides affected ADP-induced platelet aggregation even at subtoxic concentrations. These results should be taken into account when developing a new generation of antibacterial agents based on the CAMPs of Hirudo medicinalis.
